A portable isothermal amplification device with a portable heat source and a thermal sensor linking to a cloud environment via mobile devices with a mobile application of the present invention for determining the DNA information of samples with a given primer in a manner of point-of-collection (in the field). The cloud environment with mobile devices can provide isothermal amplification information that may include isothermal amplification related data (e.g., genetic information of organism, cancer cells or viruses of interest), analysis methods, solutions provided by experts for access, storage, analysis and consulting. The information may also include gene expression levels of interest, DNA identity of samples, as well as treatment suggestion and professional lists for consulting. In addition, the cloud environment can provide further solution, which is based on the sequence presence information generated from the devices for the end users.

The method may involve employing cooling exteriorly of and adjacent to the gas phase containing portion of the vessel or cooling by means of a coolant flowing within coils disposed in the interior of the vessel or both. The process is preferably a continuous process. The apparatus may be a vessel transparent to microwave energy for receiving the sample. The vessel has space overlying the liquid phase containing portion for a gas phase. Structures for cooling means for positively cooling the gas phase to reduce the pressure of the gas phase without effecting substantial cooling of the reagent are provided. These structures for cooling may be contained within the vessel, exteriorly of the vessel or modification of the vessel configuration to facilitate gas phase cooling or combinations thereof. The invention also provides a method and associated apparatus for employing microwave heating and cooling of gases evolving from the heated sample in performing chemical analysis of samples. The invention further contemplates using such a system to purify samples.

The invention relates to a device for the amplification of DNA in a reaction mixture, the device (1) comprising a heated chamber (2) including a rotor (3) for holding a plurality of reaction vessels for reaction mixtures, a drive means for the rotor, a microwave energy source (8) with means for controlled delivery of said energy to the reaction mixtures, and a system (14-16) for determining denaturation of double-stranded DNA. The invention also provides a method for the amplification of a nucleic acid strand. In the first step of the method, a reaction mixture is formed comprising the target nucleic acid strand, nucleotides, a primer, a thermostable nucleic acid polymerase, and, if necessary, a reagent for the detection of denaturation of double-stranded DNA. In the second step, the mixture is incubated at a temperature which allows synthesis of a nucleic acid strand complementary to the target nucleic acid strand. The third step comprises denaturing double-stranded DNA formed in the second step by microwave energisation of the reaction mixture with monitoring of the mixture to determine the denaturation end point. The reaction mixture is allowed to cool to a temperature at which primer anneals to the target nucleic acid strand in the fourth step. The second to fourth steps are repeated until a desired level of amplification is achieved.

An apparatus (1) for activation of chemical-physical processes comprising a microwave source (40), for example a magnetron or a klystron or a solid state oscillator (FET transistor), operatively connected at one end of a microwave antenna, for example a co-axial antenna (3), at a connector (13). The apparatus, furthermore, comprises a generator of electric current (45) at high frequency connected to an ultrasonic transducer (2) for example by a shielded cable, not shown in the FIG. In operative conditions, the antenna (3) and the ultrasonic transducer (2) are arranged substantially at right angles and at direct contact with a reacting mass (15) contained container (10) in order to activate it and then to complete a desired chemical and/or physical process. More in detail, the reacting mass (15) is subject contemporaneously, or alternatively, to an electromagnetic field generated by the antenna (3), diagrammatically indicated (30), and from the ultrasonic field (20) generated by the ultrasonic transducer (2). By arranging the microwave antenna (3) and the ultrasonic transducer (2) in the reacting mass (15) the efficiency of the process is optimized achieving a synergistic effect thanks to the combination of the electromagnetic field and the ultrasonic field.
