Disclosed is a compound having a GPR40 agonistic activity and useful for use in a pharmaceutical composition or useful as an insulin secretion promoter or a prophylactic or therapeutic agent for diabetes. Extensive studies have been made on compounds having a GPR40 agonistic activity. As a result, it is found that a compound (I) having a carboxylic acid bound to the second or third ring moiety through a methylene group or having a benzene ring bound to the second or third ring moiety through a group –O-methylene -NH-methylene, wherein the benzene ring has been substituted by a 6-membered monocyclic aromatic ring, or a pharmaceutically acceptable salt of the compound (I) has an excellent GPR40 agonistic activity. It is also found that the compound has an excellent insulin secretion-promoting activity and can prevent the increase in a blood glucose level after glucose tolerance aggressively.

[Problem] The present invention has an object to provide a compound having a GPR40 agonistic activity, which is useful as a pharmaceutical composition, an insulin secretion promoter, or an agent for preventing/treating diabetes. [Means for Solution] The present inventors have extensively studied a compound having a GPR40 agonistic activity, and as a result, they have found that the compound (I) of the present invention or a pharmaceutically acceptable salt thereof, in which a carboxylic acid is bonded to a bicyclic or tricyclic moiety through methylene, and further, a benzene ring substituted with a monocyclic 6-membered aromatic ring is bonded to a bicyclic or tricyclic moiety through -O-methylene or -NH-methylene, has an excellent GPR40 agonistic activity. They have also found that the compound has an excellent insulin secretion promoting action and strongly inhibits increase in the blood glucose after glucose loading, thereby completing the present invention.

Decachlorooctahydro-1,3,4-metheno-2H-cyclobuta(e,d) pentalen-2-one (I) is prepared by reacting SO3 with hexachlorocyclopentadiene in a mole ratio of 1.5 and 2 in the presence of Sb (III), Sn (V) halides, oxides or oxyacids as catalyst at 95-120 degrees C, and 50 to 300 psig. The reaction product is hydrolysed with 5 parts wt. aq. caustic alkali at pH >=9 at 85-115 degrees C for >=30 minutes, the pH adjusted to 5-6, with mineral acid, allowed to stand at 85-105 degrees C for 1 hour and the precipitated product (I) covered. (I) is an insecticide, particularly for controlling ants. and roaches. (I) is produced in 95% yields.

The process for producing alicyclic monoketones (hydroxyphenylcyclohexanone derivatives) according to the present invention comprises hydrogenating substituted bisphenols such as bisphenol A in a solvent in the presence of a palladium/alkali metal catalyst in which palladium and an alkali metal are both supported on a carrier to obtain alicyclic monoketones such as 2-(4-oxocyclohexyl)-2-(4-hydroxyphenyl)propane. The process for producing alicyclic diketones according to the present invention comprises hydrogenating substituted bisphenols such as bisphenol A in a solvent in the presence of a palladium/alkali metal catalyst in which palladium and an alkali metal are both supported on a carrier to obtain alicyclic diketones such as 2,2-bis(4-oxocyclohexyl)propane and 4,4′-bicyclohexanone. The other process for producing alicyclic monoketones according to the present invention comprises hydrogenating biphenols such as bis(4-hydroxyphenyl) in an organic solvent in the presence of a palladium catalyst in which 10 to 30% by weight of palladium is supported on a carrier to obtain alicyclic monoketones such as 4(4′-hydroxyphenyl)cyclohexanone. According to the present invention, alicyclic monoketones or alicyclic diketones can be obtained with high selectivity and in high yields through simple steps, because the process includes only one reaction step to hydrogenate substituted bisphenols under relatively mild conditions.
