The invention discloses a responsive nano preparation co-loaded with artesunate and chloroquine and a preparation method of the responsive nano preparation. The nano preparation is mainly formed by assembling a carrier material HPA, artesunate and chloroquine, wherein the carrier material HPA is synthesized by 4-hydroxymethyl phenylboronic acid and PLGA (poly (lactic-co-glycolic acid)). According to the nano preparation prepared by the invention, the encapsulation efficiency of the artesunate can be remarkably improved by utilizing the hydrogen bond acting force between the phenylboronic acid and the artesunate, the drug release is promoted by utilizing the ROS responsiveness of the phenylboronic acid, and the combined treatment effect of the artesunate and chloroquine drugs is exerted.
