A method for synthesizing nucleic acid under constant temperature, comprising the following steps: 1) providing a nucleic acid, the 3' end of the nucleic acid has an Flr region, the Flr region anneals with an Flc region on the same strand, and a helix ring may be formed by simultaneous annealing of the Flr and Flc regions; 2) using the nucleic acid in step 1) as a template to synthesize a complementary strand thereof; 3) performing complementary strand synthesis by means of a polymerase-catalyzed strand substitute complementary strand synthesis reaction to replace the synthesized complementary strand in step 2); wherein the nucleotide sequence of the oligonucleotide at the 5' end of the primer is substantially the same as a region synthesized by using the primer as a starting point.
