Provided are a method and kit for synthesizing a nucleic acid under constant temperature conditions, the method comprising: 1) providing a nucleic acid, wherein the 5'-terminus of the nucleic acid has an Nc region that can be annealed to the N region on the same chain, while the 3'-terminus of the nucleic acid has an Nc region that can be annealed with the N region on the same chain, and the Nc regions of the 5'-terminus and the 3'-terminus of the nucleic acid have a competitive relationship against annealing with the N regions on the same chain; 2) using the nucleic acid described in step 1) as a template and the 3'-terminus of the Nc region that has been annealed with the N region as a starting point of synthesis to synthesize its own complementary strand; and 3) carrying out the complementary strand synthesis by means of a polymerase catalytic strand displacement type complementary strand synthesis reaction to replace the complementary strand synthesized in step 2).
