ABSTRACT OF THE DISCLOSURE The present invention relates to a method for pre-paration of ion exchangers from hydrophilic anion-exchanging gels based on acrylate or methacrylate copolymers prepared by copolymerization of amino-group-containing monomers, selected from a group comprising aminoalkyl acrylates and methacrylates, aminoalkylacrylamides and aminoalkylmethacrylamides or on gels prepared by chemical bonding of amine groups to the hydro-phylic matrix of acrylate or methacrylate copolymers. The me-thod consists in alkylation or hydroxyalkylation of amine groups by alkylating agents, as alkyl halogenides, alkyl p-toluenesul-fonates, alkylsulfates and epoxides. Alkylation with alkyldiazo compounds was used for transformation of the undesired carboxy-lic groups of the ion exchangers. Another modification consists in the reaction of halogenated acrylate or methacrylate gels with polyamines or oligomeric polyethylene imines. These reac-tions lead to ion exchangers with modified chemical nature, dis-sociation constant, exchange capacity, degree of hydrophility, distance of the ionogenous groups from the surface of the matrix or the character of microsurroundings. The resulting ion-exchan-ging materials are suitable above all for the sorption and se-paration of biopolymers.

Ethylenically unsaturated monomers are polymerized with an anionic initiator by adding the initiator portionwise to a stirred polymerizing mixture throughout the course of reaction and effecting the additions only when the temperature of the reactants falls below that temperature at which the effectiveness of the initiator is decreased to one half in 15 minutes or less under the conditions of the reaction. The reaction may be carried out in bulk or in a diluent, and may be terminated at any desired degree of conversion. Exemplified initiators are tert.-butyl lithium, potassium tert.-butyl alcoholate, lithium tert.-butyl alcoholate, sodium tert.-butyl alcoholate, and triphenylmethyl sodium. Exemplified monomers are methyl methacrylate, butyl methacrylate, methyl acrylate, butyl acrylate and acrylonitrile. The Specification includes a drawing of a suitable reactor.

Die Erfindung beschreibt ein einfaches Chromatographieverfahren zur Gewinnung und Reinigung des Plasminogenaktivators Urokinase. Erfindungsgemaess werden urokinasehaltige Loesungen bei schwach saurem bisneutralem p H-Wert mit neuen Cyanoalkylgruppen enthaltenden Traegern vom Methacrylattyp in Kontakt gebracht. Dabei wird die Urokinase durch Adsorption an die Traeger aus den Loesungen abgetrennt. Durch Desorption von den Traegern mit Loesungen alkalischen p H-Wertes, Dialyse und Lyophilisation wird die Urokinase in reiner Form erhalten. The invention describes a simple chromatography method for obtaining and purifying the plasminogen activator urokinase. According to the invention, urokinase-containing solutions are brought into contact with novel cyanoalkyl group-containing methacrylate-type carriers at a weakly acidic bisneutral p H value. The urokinase is separated from the solutions by adsorption to the carriers. By desorption of the carriers with solutions of alkaline pH value, dialysis and lyophilization, the urokinase is obtained in pure form.

The density of crosslinking is controlled above all by variation of the concentration of the initial dextran solution and the re-sulting gel is optionally subjected to the additional crosslinking in a separate subsequent operation. The pro-cedure according to the invention provides the product of higher quality with a lower content of carboxylic groups and substantially facilitates the preparation of gels with a very high and a very low crosslink density.

The storage container of samples for analysis according to the invention finds the application in general and clinical analyses, toxicology, environmental protection, agriculture, food industry, biology and biotechnologies for entrapping, storage, preparation and processing of real samples after withdrawing from a source and before the proper analytical determination. The design of the storage container of samples substantially reduces time of sample processing at users and also substantially reduces demands for material and labour in manufacturing.

Saccharides and their derivatives are selected from the group comprising monosaccharides, oligosaccharides, deoxy sugars, amino saccharides, acylated saccharides, ether or halogen derivatives of monosaccharides and oligosaccharides, and the like. The invention also pertains to the application of these macroporous three dimensional copolymers with saccharides as sorbents and materials for the affinity chromatography for isolation of physiologically effective materials which contain free bonding sites for saccharides and are able to form specific complexes with saccharides.

The compound containing two or more reactive groups per molecule is preferably cyanogen bromide, epichlorohydrin, a diepoxide, a polyepoxide, a diisocyanate or a polyisocyanate. Preferred anion and cation forming compounds and mixtures are an amino acid, a diamine and an amino acid or hydroxyacid, or a dicarboxylic acid and a diamine. In the examples, copolymers of 2-hydroxyethyl methacrylate and ethylene glycol diacrylate; 2- hydroxyethyl acrylate and methylene-bis-acrylamide; 2-hydroxyethyl methacrylate and ethylene glycol dimethacrylate; and diethylene glycol mono methacrylate and ethylene glycol diacrylate; are reacted with (a) epichlorohydrin and arginine; (b) cyanogen bromide and arginine; (c) diepoxybutane and arginine; (d) hexamethylene diisocyanate and arginine; (e) diepoxybutane and taurine and (f) diepoxybutane and epsilon amino caproic acid.

ŘeSení se týká způsobu kvantitativního fluorimetrického stanovení peroxidu vodíku a substrátů oxidáz. Tento způsob spočívá v injekci vzorku substrátu do kontinuálního proudu média. Stykem média s imobilizovaným enzymem odpovádajícím přislužnému substrátu vzniká při enzymatické oxidaci substrátu peroxid vodíku. Ten přichází dále do styku s lmohllizovanou peroxidázou. Médium obsahuje chemickou látku, která v přítomnosti peroxidu vodíku a peroxidázy je schopna vytvořit fluorescenční produkt. Vykazovaná fluorescence Je měřena fluorimetrem. Samotná kvantifikace se provádí na základě porovnáni íluorimetřické odezvy na stanovený vzorek substrátu a odezvy na známá standardní množství zkoumané látky.
