Conformational Epitope Initiated Signal Amplification

This invention relates to a method to generate a signal used to detect the presence or quantity of a biomarker in a sample. The signal generating reaction is initiated when the biomarker under assay interacts with a specific binding partner. The interaction produces a structural change in the binding partner that is recognized by additional binding partners capable of generating a signal. The reaction produces a localized cluster of signaling molecules that can be detected above background. The signaling cluster is detectable within minutes when interrogated in a chamber of specific dimensions. The presence of the signaling clusters is a qualitative indication of the presence of the analyte, while the number of signaling clusters detected is a direct quantification of the number of biomarker molecules in the sample. The reaction can be formatted to detect proteins, nucleic acids, cells or other informative biomarkers.

A footwear upper comprises a front section and a rear section. The front section is at least partially forward of the rear section and at least partially defines a foot-receiving cavity. The front section has a medial edge and a lateral edge partially defining a foot entry opening. A medical device comprising radiopaque water-dispersible metallic nanoparticles, wherein the nanoparticles are released from the medical device upon implantation of the device. The medical device of the present invention is sufficiently radiopaque for x-ray visualization during implantation, but loses its radiopacity after implantation to allow for subsequent visualization using more sensitive imaging modalities such as CT or MRI. The nanoparticles are formed of a metallic material and have surface modifications that impart water-dispersibility to the nanoparticles. The nanoparticles may be any of the various types of radiopaque water-dispersible metallic nanoparticles that are known in the art. The nanoparticles may be adapted to facilitate clearance through renal filtration or biliary excretion. The nanoparticles may be adapted to reduce tissue accumulation and have reduced toxicity in the human body. The nanoparticles may be applied directly onto the medical device, e.g., as a coating, or be carried on the surface of or within a carrier coating on the medical device, or be dispersed within the pores of a porous layer or porous surface on the medical device. The medical device itself may be biodegradable and may have the nanoparticles embedded within the medical device itself or applied as or within a coating on the biodegradable medical device. The nanoparticles may be released by diffusion through the carrier coating, disruption of hydrogen bonds between the nanoparticles and the carrier coating, degradation of the nanoparticle coating, degradation of the carrier coating, diffusion of the nanoparticles from the medical device, or degradation of the medical device carrying the nanoparticles.

The computer then calculates a satisfaction function for each of these terms based on all of the individual inputs. Next, the computer calculates a joint satisfaction function for all of the terms as a function of the particular relevant decision variable, and then calculates the mutual satisfaction function for the offering party and the other parties, also as a function of the particular relevant decision variable. Finally, the computer calculates the set of decision variable yielding the maximum mutual satisfaction and provides this output to the parties.

PERIODONTAL PROBE SYSTEM ABSTRACT A periodontal probe system for measurement, storage, and display of periodontal pocket depth, gingival level and periodontal attachment level of the teeth. In the system, a periodontal probe instrument includes an elongate tip with a measurement arrangement that simultaneously measures periodontal pocket depth, gingival level, and periodontal attachment level at each of a plurality of probe sites around a tooth. The measurement arrangement provides depth signals indicative of the pocket depth and attachment level measured. Gingival level is computed as the arithmetic difference between the attachment level and the pocket depth. A pressure sensor in the probe provides a pressure signal indicative of the pressure acting on the tip of the probe. A signal processing apparatus is connected to receive the pressure signal and the depth signals. A periodontal measurement is made by inserting the tip of the probe into a gum pocket and gradually increasing the pressure on the probe tip after insertion. At a predetermined pressure level, the signal processing apparatus samples the depth signals and converts them into measurement signals corresponding to pocket depth and attachment level. The signal processor accumulates measurement signals in an examination file format. A display apparatus responds to converted depth signals by displaying pocket depth, gingival level, and attachment level for the tooth being measured in a dental chart format. In the primary embodiment, the measurement arrangement [ORILPA.H17] includes an array of opto-electronic elements disposed on the probe tip and an optical shield slidably disposed on the probe tip over the array of opto-electrical elements. When the probe tip is inserted into a gum pocket, the optical shield engages the crown of the tooth, serving as a reference point while the tip is moved into the pocket. Said modifier has a low viscosity at ambient temperatures and is a low-molecular weight urethane adduct formed by the reaction of (1) at least one polyisocyanate and at least one monohydric alcohol or (2) at least one monofunctional isocyanate with at least one monohydric or polyhydric alcohol or (3) at least one isocyanate-terminated quasi-prepolymer with at least one monohydric alcohol. Said modifier can also consist of mixtures of (1), (2), and (3). Utilizing halogenated alcohols, flame-retardant polymer foams such as polyurethane foams can be prepared. Unexpectedly, microcellular, rigid, high-resilience, and flexible foamed polyurethanes can be prepared utilizing substantial proportions, such as flame-retardant amounts, of the modifier compound without substantial reduction in physical properties of the foam.

A flushable catheter assembly includes a catheter shaft having proximal and distal end portions. The catheter may include a funnel assembly associated with the distal end portion of the catheter shaft and/or an introducer cap assembly associated with the proximal end portion of the catheter shaft. A groove is defined along at least a portion of an outer perimeter of the funnel assembly or introducer cap assembly, with the groove being configured to receive at least a portion of the catheter shaft for wrapping the catheter shaft around the outer perimeter when disposing of the catheter. Instead of (or in addition to) a groove, one or more channels may pass through the funnel assembly to receive a portion of the catheter shaft for securing the catheter shaft to the funnel assembly for disposal. The catheter assembly and/or its individual components may be formed of a water disintegrable material.

Coin-controlled apparatus for locking shopping carts together in nested series at a cart parking station having a coin-controlled mechanism mounted on a cart, e.g. on the handle of the cart, which is adapted for receiving and releasably locking therein a latch bar on a tether which is attached to the next cart in the nested series, requiring deposit of a coin to release the latch bar to free the cart for being wheeled away by the user, and holding the coin until the user brings the cart back to a cart parking station, nests it in the end cart at the parking station, and inserts the latch bar which is tethered to said end cart in the mechanism to lock the returned cart to the series and to provide for return of the user's coin.
