The method includes purifying the nucleic acid after it has been immobilized on a magnetically-responsive solid support. A pipette of the analyzer is used to form a reaction mixture comprising the purified nucleic acid and all reagents required to perform a nucleic acid amplification. Amplification products are synthesized that include a nucleotide sequence contained in the nucleic acid or the complement of the nucleic acid. The amplification products are exposed to a probe in a mixture, where the probe forms a hybrid with one of the amplification products. The formation of the hybrid in the mixture provides an indication of the presence of the nucleic acid in the sample.

(and preferably 50 g.) is produced at the centre of the holder 25. The rotational axis of the holder intersects the vertical at an angle of 15-70 degrees, (preferably 55 degrees to the horizontal) and the rotors are driven in opposite directions at a speed ratio between 3 : 1 and 10 : 1 and at such speed that all parts of the holder 25 have a resultant angular velocity about the vertical rotary axis in the same direction as the rotor 15. The rotor 15 has an adjustable counterbalance 23; the mixing time may be controlled by a timer 45; there may be a single driving motor 30, or separate motors; the apparatus may be secured to a bench by a suction pad 48. With a main rotor speed of 2,300 rev./min. and holder speed of 500 rev./min. and a radius of 1À5 inch from the axis of the rotor 15 to the centre of the mixing vessel bottom, a centrifugal field of the order of 100-200 g. is obtained. The time taken to accelerate to full speed is about 3 secs. and the total mixing time 5-25 sees. Centrifugal casting.-If the mixing vessel have a suitable interval profile and the process be continued until the materials harden, they may be removed as a moulded mass.

A method for performing an automated diagnostic assay includes an automated process for isolating and amplifying a target analyte. The process is performed by automatically moving each of a plurality of reaction receptacles containing a solid support material and a fluid sample between stations for incubating the contents of the reaction receptacle and for separating the target analyte bound to the solid support from the fluid sample. An amplification reagent is added to the separated analyte after the analyte separation step and before a final incubation step.
