The invention provides a preparation method and application of a novel Fc fusion nano antibody CTLA-4 Nb16-Fc, the structure of the CTLA-4 Nb16-Fc comprises a first functional domain of an Fc segment capable of prolonging the plasma half-life period and mediating the cytotoxic effects of ADCC, ADCP, CDC and the like, and a second functional domain capable of specifically binding and effectively blocking CTLA-4 molecules on the surfaces of T cells; and the first functional domain and the second functional domain are sequentially connected from the N end to the C end. The invention also provides a preparation method and application of the CTLA-4 Nb16-Fc fusion protein nano antibody. The novel fusion nano antibody provided by the invention can eliminate an immunosuppression function in tumor immunotherapy, and also can exert additional Fc-mediated ADCC, ADCP, CDC and other cytotoxic effect functions at the same time.
