The present invention provides a method for preparing a multilayer cell sheet through a three-dimensional single culture process by incorporating cardiac stem cells into a hydrogel, using a biodegradable natural polymer hydrogel as a three-dimensional support. According to the present invention, the multilayer cell sheet does not require special equipment to manufacture; has good physical properties, and thus can be easily handled; increases the survival rate of cells after transplantation due to having various growth and protection factors and extracellular matrix sufficiently accumulated between the cells; and facilitates movement of nutrients since the thickness thereof is thin due to cell-induced contraction.
