The present invention relates to a biosensor using a field effect transistor. The biosensor includes: a substrate; a source and drain which are disposed on the substrate and spaced apart from each other with a channel therebetween; a floating gate and control gate which are insulated from the channel by an insulation layer; and a biomaterial layer formed on the floating gate. According to the present invention, the biomaterial layer may be attached to the floating gate of the field effect transistor to provide a structurally stable biosensor. According to the present invention, since the channel of the field effect transistor is prevented from being directly exposed to a buffer solution contacting the biomaterial, variation in the characteristics of the biosensor as well as dispersion of measured values and the misjudgment rate due to the characteristic variation of the biosensor may be reduced.
