The present invention relates to a method for preventing, alleviating, and treating amyotrophic lateral sclerosis (ALS). According to the present invention, the ALS is associated with damage to an ubiquitin proteasome system (UPS), and UPS is closely associated with TDP-43-ATXN2. The present invention also identified PTK2 as an important mediator of neurotoxicity caused by damage to UPS in an ALS model by using Drosophila and mammalian cell models. The present invention demonstrates that modulating PTK2 activity can have an advantageous effect on preventing, alleviating, and treating the ALS.
