... 본 발명은 소금과 자당을 이용한 염 침출법을 통해 조직 및 뼈 재생을 위한 실크 피브로인 (또는 실크 단백질) 다공성 3차원 지지체의 제작 방법에 관한 것이다. 조직공학은 생체 내 결손된 장기 및 조직의 재생을 위해 적절한 지지체를 이식하거나 조직 세포를 체외 배양하여 실제 장기나 조직과 유사한 구조와 기능을 가지는 인공 조직을 이식하여 인체 기능의 복원을 목적으로 한다. 따라서 생체재료를 조직 공학용 지지체로 활용하기 위해서는 다공성 구조를 형성해야 한다. 이는 생체재료 자체에 적절한 공극을 지니면 세포의 생장에 필요한 공간을 제공할 수 있으며 공극이 서로 연결되어 개방된 통로를 만들면 세포간 영양분, 대사 물질 및 가스 교환이 원활하게 된다. 또한 성공적인 조직의 유지 및 복원을 위해서는 만들어진 생체재료에 세포의 부착, 증식, 세포외 기질 형성과 동시에 생체 재료를 분해하는 속도 조절이 중요하다. 실크 피브로인은 누에고치에서 세리신을 제거 한 천연 고분자로서 대량 생산이 가능한 단백질이며 FDA에서 안전성이 입증되었고, 다른 합성 고분자들과는 다르게 체내에서 면역반응이 일어나지 않고 염증반응이 적어 생체적합성이 우수하며 결정화도에 따라 분해 속도를 조절 할 수 있어 생화학 및 의학적 응용이 활발한 의료용 소재이다. 이러한 실크 피브로인을 이용해 3차원 다공성지지체를 제작하는 방법으로는 캐스팅 방법, 전기방사, 염 침출법 등이 있다. 그 중 염 침출법에 관한 연구는 활발히 진행되어왔으나, 실크 피브로인 수용액을 이용하여 지지체를 제작하기에는 기계적 강도가 약하고, 염 제거 후 동결건조 과정을 거쳐 제작 시간이 오래 걸리는 등의 제한점이 있다. 본 발명에서는 이러한 단점을 보완하여 기계적 물성을 강화하고, 연골 조직의 분화와 골 재생 효과가 우수한 지지체를 제작하였다. 따라서 본 발명에 따른 제조방법으로 제작된 실크 피브로인다공성 3차원 지지체는 조직 재생용 지지체 뿐만 아니라 정형외과적 인공 뼈, 치주 조직 재생 유도재 및 골 이식재 등으로 활용될 수 있다.
