The present invention discloses a S-pantoprazole sodium preparation method, which comprises: (1) carrying out a reaction of a chiral resolution reagent S-1,1,2-triphenyl-1,2-ethanediol and racemic pantoprazole in a C1-C4 alcoholic solution, and carrying out cooling crystallization to obtain a S-pantoprazole and S-1,1,2-triphenyl-1,2-ethanediol complex; and (2) carrying out dissociation on the complex with an alkali, removing the chiral resolution reagent, neutralizing with an appropriate acid to obtain S-pantoprazole, and carrying out a reaction of the S-pantoprazole and a sodium salt to generate the S-pantoprazole sodium salt. According to the present invention, the method has characteristics of high yield, simple process operation, easy solvent recovery, and low cost, and is suitable for industrial production, wherein the proportion of the obtained isomer is up to more than 99.8%.
