The method comprises the following steps of: inoculating a pre-cultured explant into bacterial liquid of an agrobacterium strain EHA105 with a binary plasmid p3300 for impregnation; co-culturing the impregnated explant, and screening in a screening culture medium I and a screening culture medium II which contain glyphosate; performing regeneration culturing and inducing in a regeneration culture medium I and a regeneration culture medium II sequentially to form embryoid and a seedling body; selecting the seedling body for pot culture to obtain a resistant plant; and verifying and screening by polymerase chain reaction (PCR) detection and a gel electrophoresis method to obtain a positive plant serving as a transgenic plant. In the method, the glyphosate is used as a screening agent to perform resistance screening on the Bar gene, so that the screening efficiency is improved greatly.
