The invention relates to a method for creating male sterile lines in plant by genetic engineering. According to the structural and functional characteristics of the Ssp DnaE intein internal peptide and Barnase toxoprotein, the method is used to construct a self propagation line A and line B; and the hybrid filial generation shows sterility through the hybridization between the line A and the line B. After the hybridization of the sterile line and any other non-transgene fertile line, the fertility of the hybrid filial generation is restored. The method solves the problem that the fertility of a sterile line obtained by a conventional genetic engineering method cannot be restored by a simple method.
