4D temporal data structures for visualization of alternative mission plans over time

A non-transitory computer readable medium having computer executable instructions stored thereon is provided. The computer executable instructions include a mission definition file (MDF) that includes mission data that describes alternative routes generated by at least one mission planner for a vehicle. An MDF parser extracts the mission data for the alternative routes from the MDF and maps the mission data into a temporal data structure describing the alternative routes. The temporal data includes a route list to describe the alternative routes for the vehicle and a route segment list to describe an estimated time between each of the waypoints for each segment of the route list. A waypoint list describes a latitude, a longitude, and an altitude for each of the waypoints and a mission plan constructor generates a network topology of a mission plan depicting the alternative routes for the vehicle over time.

Pathways Underlying Pancreatic Tumorigenesis and an Hereditary Pancreatic Cancer Gene

There are currently few therapeutic options for patients with pancreatic cancers and new insights into the pathogenesis of this lethal disease are urgently needed. To this end, we performed a comprehensive analysis of the genes altered in 24 pancreatic tumors. First, we determined the sequences of 23,781 transcripts, representing 20,583 protein-encoding genes, in DNA from these tumors. Second, we searched for homozygous deletions and amplifications using microarrays querying ˜one million single nucleotide polymorphisms in each sample. Third, we analyzed the transcriptomes of the same samples using SAGE and next-generation sequencing-by-synthesis technologies. We found that pancreatic cancers contain an average of 63 genetic alterations, of which 49 are point mutations, 8 are homozygous deletions, and 6 are amplifications. Further analyses revealed a core set of 12 regulatory processes or pathways that were each genetically altered in 70% to 100% of the samples. The data suggest that dysregulation of this core set of pathways is responsible for the major features of pancreatic tumorigenesis. 165-. (canceled)66. Human cancer is caused by the accumulation of mutations in oncogenes and tumor suppressor genes. To catalogue the genetic changes that occur during tumorigenesis, we isolated DNA from 11 breast and 11 colorectal tumors and determined the sequences of the genes in the Reference Sequence database in these samples. Based on analysis of exons representing 20,857 transcripts from 18,191 genes, we conclude that the genomic landscapes of breast and colorectal cancers are composed of a handful of commonly mutated gene “mountains” and a much larger number of gene “hills” that are mutated at low frequency. We describe statistical and bioinformatic tools that may help identify mutations with a role in tumorigenesis. These results have implications for understanding the nature and heterogeneity of human cancers and for using personal genomics for tumor diagnosis and therapy. 1. Medulloblastoma genes as targets for diagnosis and therapeutics

Medulloblastoma (MB) is the most common malignant brain tumor of children. To identify the genetic alterations in this tumor type, we searched for copy number alterations using high density microarrays and sequenced all known protein-coding genes and miRNA genes using Sanger sequencing. We found that, on average, each tumor had 11 gene alterations, markedly fewer than in common adult cancers. In addition to alterations in the Hedgehog and Wnt pathways, our analysis led to the discovery of genes not previously known to be altered in MBs. Most notably, inactivating mutations of the histone H3K4 trimethylase genes MLL2 or MLL3 were identified in 16% of MB patients. These results demonstrate key differences between the genetic landscapes of adult and childhood cancers, highlight dysregulation of developmental pathways as an important mechanism underlying MBs, and identify a role for a specific type of histone methylation in human tumorigenesis.

Consensus Coding Sequences of Human Breast and Colorectal Cancers

Analysis of 13,023 genes in 11 breast and 11 colorectal cancers revealed that individual tumors accumulate an average of ˜90 mutant genes but that only a subset of these contribute to the neoplastic process. Using stringent criteria to delineate this subset, we identified 189 genes (average of 11 per tumor) that were mutated at significant frequency. The vast majority of these genes were not known to be genetically altered in tumors and are predicted to affect a wide range of cellular functions, including transcription, adhesion, and invasion. Analysis of 13,023 genes in 11 breast and 11 colorectal cancers revealed that individual tumors accumulate an average of ˜90 mutant genes but that only a subset of these contribute to the neoplastic process. Using stringent criteria to delineate this subset, we identified 189 genes (average of 11 per tumor) that were mutated at significant frequency. The vast majority of these genes were not known to be genetically altered in tumors and are predicted to affect a wide range of cellular functions, including transcription, adhesion, and invasion. These data define the genetic landscape of two human cancer types, provide new targets for diagnostic and therapeutic intervention and monitoring. 1. Analysis of 13,023 genes in 11 breast and 11 colorectal cancers revealed that individual tumors accumulate an average of ˜90 mutant genes but that only a subset of these contribute to the neoplastic process. Using stringent criteria to delineate this subset, we identified 189 genes (average of 11 per tumor) that were mutated at significant frequency. Genomic Landscapes of Human Breast and Colorectal Cancers

Human cancer is caused by the accumulation of mutations in oncogenes and tumor suppressor genes. To catalogue the genetic changes that occur during tumorigenesis, we isolated DNA from 11 breast and 11 colorectal tumors and determined the sequences of the genes in the Reference Sequence database in these samples. Based on analysis of exons representing 20,857 transcripts from 18,191 genes, we conclude that the genomic landscapes of breast and colorectal cancers are composed of a handful of commonly mutated gene “mountains” and a much larger number of gene “hills” that are mutated at low frequency. We describe statistical and bioinformatic tools that may help identify mutations with a role in tumorigenesis. These results have implications for understanding the nature and heterogeneity of human cancers and for using personal genomics for tumor diagnosis and therapy. 1. An assembly for a fire fighter self-contained breathing apparatus , comprising:a frame having a user support portion mounted to the frame; anda compressed gas cylinder mounted to the frame, the single cylinder including a pressure volume portion for containing a volume of gas pressurized to a service pressure of between about 5,000 and 6,000 psig, the pressure volume portion having a length, a diameter, and a water volume;wherein the cylinder further includes a gas transmission port coupled to a pressure regulator assembly; 'wherein at least one of a length and a diameter of the pressure volume portion is at least slightly less than at least one of a corresponding length and diameter of a compressed gas cylinder for a self-contained breathing apparatus having a service pressure of about 4500 psig.', 'wherein the pressure volume portion defines an operational parameter of the cylinder, the operational parameter being a relationship between free air capacity of the cylinder in liters and a rated service time in minutes; and'}20. Consensus Coding Sequences of Human Breast and Colorectal Cancers

Analysis of 13,023 genes in 11 breast and 11 colorectal cancers revealed that individual tumors accumulate an average of ˜90 mutant genes but that only a subset of these contribute to the neoplastic process. Using stringent criteria to delineate this subset, we identified 189 genes (average of 11 per tumor) that were mutated at significant frequency. The vast majority of these genes were not known to be genetically altered in tumors and are predicted to affect a wide range of cellular functions, including transcription, adhesion, and invasion. These data define the genetic landscape of two human cancer types, provide new targets for diagnostic and therapeutic intervention and monitoring. 1. An automated system for storing and retrieving objects from multiple object categories. A plurality of storage rack assemblies is included. Each storage rack assembly has a plurality of slots sized for storing objects. A storage transport is movable alongside each respective storage rack assembly for positioning adjacent any slot. A plurality of horizontal tracks are arranged to be perpendicular to the storage rack assemblies. Each storage rack assembly has an end adjacent the tracks. A runner transport is coupled to and movable on each track for receiving one of the objects from, or providing one of the objects to a storage transport. Two conveyor rack assemblies are located at opposite ends of the tracks, aligned perpendicularly to the track. Conveyor transports are adjacent the conveyor rack assemblies for transferring objects between the conveyor rack assembly and the runner transports. Storage and retrieval operation requests are generated by simultaneously providing positioning signals to a conveyor transport, a runner transport and a storage transport. Storage transports are selected in accordance with a predetermined transport selection function. Storage slots are selected in accordance with a predetermined slot selection function. A database stores the category, age and location of each of the objects.
