The invention belongs to the field of genetic engineering, in particular relating to a site-specific recombinase, process for preparation and application. The invention provides a novel site-specific recombinase of which the specificity identifies the below structural formula which indicates a dideoxynucleotide sequence, the dideoxynucleotide sequence is cracked in the position of arrows, and a sequence of attB recognition site and the sequence indicated by the structural formula are reconnected at the cleavage site (see figure). The site-specific recombinase of the invention is capable of specifically identifying a genome MpsL1 site with comparatively high activity, and thereby the site-specific recombinase of the invention can be an effective tool in technical operation of the genetic engineering, in particular to gene site-specific insertion recombination.
