The invention discloses a medical wheat protein film which is prepared by adding polyalcohol plasticizer and cosolvent into degreased wheat protein, and processed by the working procedures of dissolving, forming a film and fixed molding; and the obtained wheat protein film has the thickness of 0.1-5mm, takes on a transparent, semi-transparent or non-transparent state, and has the tensile strength of 0.1-12MPa. The invention also discloses a preparation method of the wheat protein film. The wheat protein film not only is soft and moderate in mechanical strength, but also has good water absorbing, water retaining and air permeability properties as well as good blood compatibility and biocompatibility; and furthermore, the medical wheat protein film has no immunogenicity, can be used for absorbing proper medicine solution for slow release of medicine, and also can be used as surgical dressing, a tissue isolating diaphragm or scaffold material of tissue engineering.
