The present invention relates to biological converting and biological catalyzing technology, and is especially screening method of phophonomycin biologically converting strain. The present invention cultures the strain to be detected in cylindrical agar block as solid culture medium, utilizes colibacillus as indicating bacterium for biological identification, and sets the agar block backwards on biological identification disc to screen phophonomycin converting strain. According to that different strains to be screened in conversion capacity and grown on isometric agar block with the same amount of precursor will produce different phophonomycin concentrations and show bacteriostasis rings of different sizes on the indicating bacterium plate, the strain with phophonomycin converting predominance may be sorted out. The method is simple and suitable for initial screening of antibiotic converting bacteria.
