A METHOD FOR GENERATING BIRNAVIRUS FROM SYNTHETIC RNA TRANSCRIPTS

A system for the generation of live Birnavirus such as infectious bursal disease virus (IBDV), a segmented double-stranded (ds)RNA virus of the Birnavirdae family, using synthetic transcripts derived from cloned DNA has been developed. Mutogenesis of the Clemson University (CU) vaccine strain of P. multocida with N-methyl-N-nitro-N-nitrosoguanidine resulted in temperature sensitive (TS) mutants which grew at 37°C but not 42°C. Two mutants, PM#1 and PM#3, were found to provide turkeys with a level of protection against challenge with a virulent serotype 3 P. multocida strain (P-1059) comparable to the protection provided by the CU strain. Intravenous (IV) inoculation of PM#1, PM#3, and CU was used to determine differences in virulence. PM#1 and PM#3 resulted in lower mortality and lameness than the CU strain. Histopathological evaluation of spleens at 24, 48, and 72 hours post IV inoculation demonstrated that the CU strain induced 2 times more splenic necrosis than either PM#1 or PM#3.

Methods for providing improved immunization against coccidiosis and other bacterial, viral, or parasitic diseases in poultry. One method includes administering a solution of i(Propionibacterium acnes) suspended in normal saline to a chick at age day one, following hatching. An anticoccidial vaccine and/or other vaccine is then administered to the chick such as by oral administration. The electron beam irradiated cells of Salmonella are effective for stimulating protective immune responses in the animals against the Salmonella . Induction of these immune responses significantly reduces or eliminates the colonization of the animal by the Salmonella , and consequently reduces or eliminates the shedding of Salmonella in the feces of the animals.

A method for generating birnavirus from synthetic rna transcripts

A system for the generation of live Birnavirus such as infectious bursal disease virus (IBDV), a segmented double-stranded (ds)RNA virus of the Birnavirdae family, using synthetic transcripts derived from cloned DNA has been developed. Independent full-length cDMA clones were constructed which contained the entire coding and noncoding regions of RNA segments A and B of IBDV, respectively. Synthetic RNAs of both segments were produced by in viro transcription of linearized plasmids with T7 RNA polymerase. Transfection of Vero cells with combined plus-sense transcripts of both segments generated infectious virus as early as 36 hours post-transfection. The development of a reverse genetics system for dsRNA viruses will greatly facilitate studies of the regulation of viral gene expression pathogenesis, and design of a new generation of live and inactivated vaccines.

A presente invenção trata de um sistema que foi desenvolvido para a geração de Birnavírus ativo, tal como o vírus da doença bursal infecciosa (IBDV), um vírus segmentado de RNA duplamente espiralado (ds) da família dos Birnaviridae, usando transcritos sintéticos derivados de DNA clonado. Os clones de cDNA independentes de inteira extensão, foram construídos contendo regiões de inteira codificação e descodificação dos segmentos A e B do RNA, do IBDV, respectivamente. Os RNAs sintéticos de ambos os segmentos foram produzidos por meio de transcrição in vitro de plasmídios linearizados com polimerase de RNA T7. A transfecção das células de Vero combinado com os transcritos mais sensíveis de ambos os segmentos gerou o vírus infeccioso logo nas 36 horas após a transfecção. O desenvolvimento de um sistema genético inverso para vírus de RNA duplamente espiralados, irá facilitar acentuadamente as pesquisas de regulação da expressão do gene viral, da patogênese e o projeto de uma nova geração de vacinas ativas e inativadas.
