... 표적분자(핵산, 지질, 당, 단백질, 펩타이드 등의 생체분자나 호르몬, 저분자 화학물질, 독성물질, 이온 등을 모두 포함)를 인식하는 DNA/RNA 압타머를 선별하기 위해서는 Systematic Evolution of Ligand Exponential Enrichment (SELEX) 과정을 수반한다. 일반적으로 SELEX를 수행하기 위해서는 표적분자를 기판이나 비드 표면에 고정시키는 과정이 필수적으로 요구된다. 이는 표적물질과 결합되는 압타머와 결합하지 않는 압타머를 효과적으로 분리할 수 있기 때문이다. 그러나 이 경우 표적분자를 화학적으로 변형시키거나 tag을 붙이는 과정과 표면이나 비드에 강하게 고정화해야 하는 과정이 필수적으로 수반되어 실제 원래의 표적물질의 결합능력이 떨어지기도 한다. 또한 SELEX과정의 각 round에서 실제 압타머 라이브로리가 표적물질과 잘 결합하고 있는지에 대해 관찰할 수 있는 positive/negative 모니터링이 불가능하기 때문에 수회에 걸친 round이후 선별된 압타머를 분석함으로써 SELEX과정이 올바르게 진행되고 있는지를 간접적으로 확인하고 있다. 이러한 종래의 문제점들을 획기적으로 해결함과 동시에 보다 손쉽고 간편한 SELEX 기법을 구성하기 위해, 본 발명에서는 금나노입자를 이용한 새로운 SELEX 기법을 제시하고자 한다. 즉 표적분자의 변형없이 원래의 표적을 그대로 사용할 수 있고 압타머 라이브로리와 표적물질의 결합력이 증가되고 있는지를 SELEX 과정에서 손쉽게 관찰할 수 있다. 이러한 방법은 고분자 물질 뿐만 아니라 저분자물질에 폭넓게 활용될 수 있을 뿐만 아니라 항체생산이 어렵거나 항체사용에 제약이 있는 표적물질을 손쉽게 인식하는 센서로서 널리 응용될 수 있다.
