인간 다능성 줄기세포로부터 유래된 심근세포를 이용한 in vitro 심장독성 평가 기술

... 본 발명은 세포 외 미세 환경을 모사할 수 있는 매트리겔(matrigel)을 기반으로, 인간 다능성 줄기세포를 심근세포(cardiomyocytes)로 분화시킨 후, 전기생리학적 방식으로 약물-유도성 심장독성을 평가하는 방법에 관한 것으로, 구체적으로 인간 다능성 줄기세포(human pluripotent stem cell; hPSCs)의 분화 시작 단계(d-3)에서 세포 외 3차원 미세 환경을 매트리겔(Matrigel)을 이용하여 구성한 후, wnt 신호를 활성 및 억제할 수 있는 소분자(CHIR99021, IWP4)를 처리하여, 심근세포 분화 성공률이 92%를 나타냈으며, 중배엽으로의 유도를 현저히 촉진하고 심근세포로의 분화를 매우 효과적으로 유도할 수 있음을 확인하였다(84%). 또한 분화를 마친 심근세포에서 패치 클램프(patch clamp) 실험기법을 통해 전기생리학적 특성을 확인하여 인간 다능성 줄기세포 유래 심근세포(hPSC-CMs)가 인체 내 심근세포와 유사할 뿐만 아니라 전기생리학적 특성 및 약물 유발성 부정맥을 효과적으로 재현할 수 있음을 확인하였고, 잠재적인 부정맥 유발성 약물들의 독성평가시험 모델로써 사용할 수 있음을 최종 확인하여, 본 발명의 인간 다능성 줄기세포로부터 유래된 심근세포는 약물-유도성 심장독성을 평가하는 방법에 사용될 수 있다.
