... 본 발명은 장 오가로이드 상태 및 ROCK 저해제 첨가시기의 복합처리를 통한 장 오가로이드의 효율적인 장기냉동동결보존방법을 제공한다. 본 발명에 따르면 생체 외에서 배양된 장 오가로이드를 단일세포 보다 온전한 오가로이드 (intact intestine organoid) 형태로 냉동동결보존 후 해동시 생존율이 높음을 확인하였다. 3개월 이상의 장기냉동동결시에는 ROCK(Rho-kinase) 저해제를 동결배지에 직접 첨가법이 전처리 및 해동 후 처리보다 생존회복율이 현저히 높음을 알수 있었다. 또한 해동 후 배양된 장 오가로이드의 특성 분석 결과 crypt-villi 구조를 가지는 in vivo intestine의 형태학적 특성이 그대로 유지함을 관찰할 수 있었다. 따라서, 본 발명에 적용된 고효율 냉동동결보존법은 오가로이드 기반 치료(organoid-based therapy)에 필수적인 많은 양의 세포를 제공함으로써 효과적인 장 질병과 손상 치료에 적용에 활용될 수 있을 것으로 사료된다.
