The virus establishment method comprises the following steps: adopting the virion of the wild HaSNPV (helicoverpa armigera single nucleocapsid nucleopolyhedrovirus) G4 strain with an fp25k gene not mutated as an experimental material; carrying out site-directed silent mutation twice by establishing a plasmid which contains the fp25k gene; and then carrying out cotransfection, fluorescent screening and plaque purification by establishing a plasmid which contains fluorescent protein to obtain a mutated G4 virus. According to the mutated G4 virus obtained through the method disclosed by the invention, a genetic factor which generates high mutation rate during the passage of a wild virus is eliminated and the stability is strong.
