A device for the addition of a compound or compound mixture to another under various reaction conditions, parallel or otherwise is disclosed. The method and apparatus offer significant advantages over conventional experimental reactors, and is especially useful for synthesizing and screening large numbers of compounds. The device generally consists of a body (10) containing one or more recessed areas (14) surrounding one or more hollow pistons (18) mounted in barrels (30) covered by a top-plate (24). The recessed areas are filled with compounds. The hollow pistons are filled with other compounds. Using gas pressure the pistons move. This movement results in the addition of the compounds contained in the recessed areas to the compounds contained in the pistons. Modifications as found for conventional experimental reactors can also be incorporated in the apparatus.

The combinatorial synthesis method uses a device equipped with a plurality of collectors to obtain a plurality of compositions having materially different properties. The first quantity of fluid reactant is reacted to form a first quantity of product composition. After collection of the first quantity of product composition is completed, the second quantity of fluid reactant is reacted to form a second amount of product composition that is materially different from the first amount of product composition. The apparatus has a nozzle 130 and a plurality of collectors 236 connected to the reactant source 120. The nozzle and the plurality of collectors move relative to each other such that the collector can be selectively arranged to receive a fluid stream exiting the nozzle. Multiple product compositions can be evaluated to determine their suitability for various applications.

The aim of the invention is to investigate the bonding of substances to target molecules. This is achieved by means of a densely packed device wherein various target molecules are bonded in a large number of sample areas. Cross-contamination and evaporation need to be minimized. The active surface of the sample areas have to be maximized. The inventive solution resides in the use of carrier plates, containing densely packed capillary structures and having a very large inner surface despite small outer dimensions. 1000 times more molecules can be bonded than on a flat outer surface of a comparable size. Cross contamination is avoided by the lack of cross links between the capillaries. Evaporation is minimized by a small outer surface. After the inner surface of the capillaries has been silanized, peptides and peptidomimetics are synthesized in a locally targeted manner. The molecular interactions of components of the substance library with active substances in a solution or a suspension are investigated by means of a local resolution optical detection method. Handling, especially cleaning and covering with substances, is carried out in a simple manner by rinsing liquids through the capillary plate.

The inventive apparatus is comprised of a means for producing cavitation in a complex reaction mixture to enhance the yield of the selected reaction product, a means for controlling the temperature of the complex reaction mixture, especially during cavitation, and a means for affecting magnetic separation of paramagnetic beads to which the selected reaction product is attached from the complex reaction mixture. In one embodiment, the compound(s) or molecule(s) is synthesized in situ, and isolated using the inventive apparatus. The apparatus finds use in the fields of solid phase organic synthesis, and for isolation and purification of a selected compound(s) or molecule (s), especially where automation is desired.

The reaction block assembly of the invention comprises a reaction block in which are disposed a plurality of reaction wells. Influent and effluent ducts open at the upper face of the reaction block adjacent each of the reaction wells. These ducts provide communication between each reaction well and a source of reagents and gases and with waste removal means for removal of effluent from the reaction wells. A membrane sheet overlies the upper face of the reaction block to define a pneumatic valve to open and close the duct openings each of the reaction wells to control the input of reagents and gases and effluent flow from the reaction wells. The reaction block assembly preferably includes a temperature control block that underlies the reaction block for control of the temperature in the reaction wells. A waste block underlies the reaction block or, if used, the temperature control block. The effluent ducts from the reaction wells communicate with the waste block for removal of reaction well effluent from the reaction block assembly. Overlying the reaction block is a top manifold plate block for delivery of high pressure gas to actuate the pneumatic valves by applying pressure against the membrane to seal the duct openings.

Various materials required are prepared in advance in the feeding device and directly fed to the reactor tube. The reaction or washing solution in the reactor tube is pumped to a waste liquid tank through the vacuum tube. Nitrogen is introduced into each reactor tube through the nitrogen tube. This device can be applied in batch-wise peptide synthesis using solid-phase methods.
