The present invention relates to a novel phosphate-dependent glucokinase with excellent stability, high temperature properties, and heat resistance, and a method for producing glucose-6-phosphate using the same. In a high temperature enzymatic reaction process using enzymes, it is possible to increase solubility of substrate [Poly(Pi)n, glucose], thereby enabling involvement of high concentration substrates. Owing to increased diffusion rate of substances, increased reaction rate, and reduced reaction time, unit productivity increases, and pollution in the process due to external microbes for high temperature enzymatic reaction processes can be minimized. Through application of heat resistance after recombination and mass-expression of enzymes of the present invention in a mesophilic microbe, selective denaturation and removal of protein derived from the mesophilic microbe, since high heat treatment becomes possible, can be conducted as well.
