In the present invention, disclosed are: a fermentation composition of Tricholoma matsutake mycelium of mixed grains, which has excellent flavor by covering a bad smell and adding fragrance of the Tricholoma matsutake, improved amount of essential amino acid and unsaturated fatty acid to have excellent nutrients, increased total amount of phenolics and aglycon isoflavone, and improved anti-oxidation activity and digestive enzyme inhibition activity; a preparation method thereof; and health functional food comprising the same.
