Broad spectrum benzothiophene-nitrothiazolide and other antimicrobials

The invention provides FIG. 1 novel antimicrobial chemical entities based on a nitrothiazolide backbone that exhibit antibacterial and antiparasitic action against a wide range of human pathogens. The new classes of compounds show extended action against Gram positive bacteria including MRSA drug resistant pathogens. In the Gram-positive organisms, they specifically target and functionally inhibit microbial attachment to surfaces and biofilm formation. In Gram-negative bacteria, including enteroaggregative E. coli strains, these compounds function as pilicides by inhibiting the assembly of pilin subunits into adhesive filaments. Several of these compounds show potent antimicrobial action against Gram positive bacteria, perhaps involving novel targets. Many of the benzothiophene derivatives exhibit antimicrobial activity in the low micrograms per ml range and in blocking biofilm formation in the nanomolar range; ranges considered are well within the range of utility as therapeutics.

The method is particularly robust to image data having low signal-to-noise ratio (“SNR”). An MRI system is used to acquire magnetic resonance elastography (“MRE”) data from a subject containing the material. Exemplary materials include lung tissue. Images are reconstructed from the MRE data and used to produce a wave image from which a spatial frequency spectrum is produced. A principal frequency of the spatial frequency spectrum is produced and used to calculate a mechanical property of the material. For example, shear stiffness may be calculated.

Methods of the invention comprise administering to a subject a St. thermophilus bacterium modified to release a biotherapeutic agent following bile exposure. Administration of the St. thermophilus bacterium promotes a desired therapeutic response. The bacterium may be modified to express and release AQ or AQR rich peptides which subsequently inhibit cellular apoptosis or reduce mucosal damage. Thus, methods of the invention find use in treating or preventing a variety of gastrointestinal disorders including C. difficile infection and antibiotic-associated diarrhea.

This testing strip contains at its opposite end a sealing mechanism for sealing engagement with a preservative pouch, and an oversized handle with a patient identifying mechanism. The preservative pouch of the present invention contains a pair of seals. The first seal, located proximally to the user, enables a preservative which is maintained within the pouch to be sealed during shipment of the pouch to the user. Thereafter, the user punctures the first seal upon placement of the reagent pad into the pouch. The sealing mechanism contained on the reagent pad then seals at the opening in the pouch to maintain the reagent pad in sealing arrangement within the pouch. At its distal end, the pouch contains a second sealing mechanism. This sealing mechanism helps protect the preservative as well as the oral sample, within the pouch during shipment from the user back to the testing laboratory.

A high energy photon source. A pair of plasma pinch electrodes are located in a vacuum chamber. The chamber contains a working gas which includes a noble buffer gas and an active gas chosen to provide a desired spectral line. A pulse power source provides electrical pulses at repetition rates of 1000 Hz or greater and at voltages high enough to create electrical discharges between the electrodes to produce very high temperature, high density plasma pinches in the working gas providing radiation at the spectral line of the source or active gas. A fourth generation unit is described which produces 20 mJ, 13.5 nm pulses into 2 π steradians at repetition rates of 2000 Hz with xenon as the active gas. This unit includes a pulse power system having a resonant charger charging a charging capacitor bank, and a magnetic compression circuit comprising a pulse transformer for generating the high voltage electrical pulses at repetition rates of 2000 Hz or greater. Gas flows in the vacuum chamber are controlled to assure desired concentration of active gas in the discharge region and to minimize active gas concentration in the beam path downstream of the pinch region. In a preferred embodiment, active gas is injected downstream of the pinch region and exhausted axially through the center of the anode. In another preferred embodiment a laser beam generates metal vapor at a location close to but downstream of the pinch region and the vapor is exhausted axially through the anode.

An intravascular material delivery dilation catheter (10) having a pair of longitudinally spaced inflatable balloons (20, 22) with a drug delivery region (26) defined therebetween. The catheter is ideally suited for use after a PTCA procedure, wherein the proximate balloon (20) seals the blood vessel while the distal balloon (22) is uniquely contoured when inflated to define fluid communication paths (60) therepast and proximate a blood vessel (54) to be treated. The distal balloon (22), when inflated, has four lobes but could also be textured. Each lobe is separated from the next by a groove, which groove in combination with the blood vessel inner wall forms a fluid communication path therebetween. Upon inflation of both balloons (20, 22) in a blood vessel (54), a medicament such as heparin can be injected, via the drug delivery region (26) between the inflated balloons (20, 22), wherein the medicament flows past the distal balloon (22) at a selected rate. Accordingly, a medicament can be injected directly to a treatment site rather than injected as a bolus dose, thus, a smaller dosage may be employed to minimize side effects. Alternatively, perfusion can be accomplished by only partially inflating the proximate balloon (20) to constrict flow therepast, or eliminating the proximate balloon (20) entirely, where the drug delivery region is disposed upstream of the contoured distal balloon (22).

An electric discharge laser (34) with fast wavelength correction. Fast wavelength correction equipment includes at least one piezoelectric drive and a fast wavelength measurement system (104) and fast feedback response times. In a preferred embodiment, equipment is provided to control wavelength on a slow time frame of several milliseconds, on a intermediate time from of about one to five milliseconds and on a very fast time frame of a few microseconds. Preferred techniques include a combination of a relatively slow stepper motor and a very fast piezoelectric driver for tuning the laser wavelength using a tuning mirror. Very fast wavelength control is provided with a piezoelectric load cell in combination with the piezoelectric driver. Preferred embodiments provide fast feedback control based on wavelength measurements, fast vibration control, active damping using the load cell and an active damping module (320), and transient inversion using feed forward algorithms based on historical burst data.

This invention relates to an apparatus and process for writing video information in the form of a frequency modulated signal upon a video disc member and for recovering the video information from the video record is described. The video disc member (10) formed by the writing apparatus is also described. The writing apparatus includes a laser (30) as a source of a laser write beam (29) and a write optical system (40) for directing the stationary laser write beam to a moving video disc member. The video disc member comprises a substrate member carrying a light responsive coating (26) at least one surface. The write optical system focuses the beam to a small spot of light approximately one micron in diameter upon the light responsive layer. The intensity of the focused spot is changeable under the control of a light intensity modulating assembly (44). This light intensity modulator changes the intensity of the write laser beam with respect to a predetermined threshold intensity sufficient to form a first type of indicia (37) in the coating. A read system (165) is employed to retrieve the video signal stored on the video disc member. This read system includes a read laser (150) for generating a read beam, and a motion control assembly for rotating and translating the video disc member relative to the read light beam. A read optical system directs the read beam to impinge upon the successively positioned specular light reflective and non-specular light reflective regions (37, 38) carried by the video disc member. The read optical system also collects the light (150) reflected from these regions. The reflected light beam is intensity modulated by the specular light reflective and non-specular light reflective regions. Skin patches (20) having a microprojection array (10), a reservoir (18) containing an antigenic agent and an immune response augmenting adjuvant, an d methods of using same to vaccinate animals (e.g., humans) is disclosed. In a preferred embodiment, the microprojection arrays (10) are composed of a photoetched and micro-punched titanium foil (14). The microprojections (12) are coated with a liquid formulation containing a vaccine antigen and an adjuvant such as glucosaminyl muramyl dipeptide, dried, and applied to skin of the animal to be vaccinated using an impact applicator. The microprojections (12) create superficial pathways through the stratum corneum to facilitate permeation of antigenic agent and adjuvant. Antigen dose and depth of penetration can be controlled. This technology has broad applicability for a wide variety of therapeutic vaccines to improve efficacy, and convenience of use.

The air permeable liquid impervious element is interposed between the chamber and the enclosed volume and is configured to preclude liquid from entering the chamber.
